Jeune adulte, il étudie la musique auprès de la concertiste classique Catherine Collard et s'initie en autodidacte à la musique pop et au jazz.En 1986, Jean-Marie Machado fait son irruption sur la scène du jazz avec un trio aux conceptions tout à fait originales (Trio Machado avec Louis et François Moutin), et un répertoire entièrement neuf, atteignant, selon Martial Solal, 'un degré de raffinement et de perfection rarement atteint dans ce domaine'. Cette formation est à l'affiche des plus grands festivals de jazz de l'été 2002 (Jazz à Vienne, Marciac, LaVillette, Orléans) et effectue un grand nombre de concerts sur les scènes nationales. En 2003, le quartet devient sextet pour interpréter une suite instrumentale de jazz d'aujourd'hui dédiée aux grands thèmes de la musique classique et traditionnelle andalouse (Andaloucia, Le chant du Monde - Harmonia Mundi , 2005)En 2003, Jean-Marie Machado crée un magnifique duo avec le saxophoniste américain David Liebman autour d'un répertoire dédié en partie au fado.
